3. SPECIMEN TREATMENT AND TEST PROCEDURE
(1)SPECIMEN TREATMENT
-Homogenize a reasonable amount of sample (e.g. 20g) with a suitable equipment (e.g. ultra turrax or mixer).
-Weigh 2 g of homogenized sample into a 50mL cryovial.
-Add 10 ml of 0.05 M PBST and 2.5ml absolute alcohol, shake for 5 min.
-Add 5ml n-hexane and shake for 2 min.
-Centrifuge (10 min / 4000 rpm) for separation, suck 10ml of the middle liquid into another 50ml cryovial.
-Add 6ml dichlormethane and 3ml ethyl acetate, shake for 5min
-Centrifuge (15 min / 4000 rpm) for separation. If there is emulsification,shake the tube and centrifuge again.
-Transfer 5ml of the underneath to the 5ml cryovial tube. If the liquid is not clear, suck off the buddy and dry it at 65°C with a mild stream of nitrogen.
-Dissolve the dried residue with 0.3 ml PBST only for QNs.
(2)TEST PROCEDURE:
Read the entire procedure carefully before performing any test. Keep the device at room
temperature before testing.
1. Prepare samples according to the step of Specimen treatment.
2. Remove the Quinolone Residue Rapid Test Card from sealed pouch.
3. Hold the dropper vertically and transfer 3 full drops of solution intospecimen well (S) of the test card, and then start the timer. Avoid trapping air bubbles in the specimen well (S). See the illustration above.
4. Wait for red bands to appear. The result should be read in approximately 3-5 minutes. Do not interpret results after 5 minutes.The test plate should be placed directly to the observer.
